Sen,

When you make a change on a GPO it will take effect immediatly. You should make sure that you replicate your domain controllers if you have more then one. But it will only take effect on the users or computer in the OU when they either logoff and log back on (for users) or reboot the machine (users and computers). The only other way around that is to run secedit /refreshpolicy user_policy /enforce
secedit /refreshpolicy machine_policy /enforce

that would update the policies on their computer. Otherwise you have to get them to reboot or something.

I think there is also a policy to tell the computer and user to refresh at an interval. However when you set that you have to get them to reboot or run the secedit.
