It depends.
If you need more security for the workstation other than what is applied from the defltwk.inf template then you will need to.
When you perform an upgrade of a Windows NT4 workstation to Windows 2000 Professional system it will not receive the template during the upgrade.
In this situation you would need to apply the basicwk.inf security file to bring the newly upgraded Windows 2000 system to a base level of configured security.
This would then make the system comparable to a clean install
